90% RPMI 1640 + 10% FBS + HFCS ("Hybridoma Fusion and Cloning Supplement"

saturated culture 1:3 to 1:10 every 2-5 days (some cells attach to the surface of the
culture flask, but can be detached by tapping the flask); seed out at ca. 0.3 x 106
cells/ml; a maximal density of 1.0 x 106 cells/ml should not be exceeded at 37C with
5% CO2 cell harvest of ca. 1.0 x 106 cells/ml
